Anti-Cancer Effect of 1,2,3,4,6-Penta-O-Galloyl-¥â-D-Glucose in B16-F0, Mouse Melanoma

Abstract
1,2,3,4,6-Penta-O-galloyl-¥â-D-glucose (PGG) is a hydrolyzed tannin isolated from plants such as Galla Rhois. This study evaluated the effects of PGG in B16-F0, mouse melanoma, and its molecular mechanism. PGG inhibited cell proliferation and induced cytotoxicity. In addition, PGG suppressed the clonogenic activity and inhibited the cell cycle by inhibiting the G1 and S phase. PGG impeded the protein expression of cyclin-dependent kinase 2 and cyclin E, which are checkpoint system-related proteins for the cell cycle. PGG also induced early and late apoptosis by increasing the protein expression of the intrinsic (cytochrome C and cleaved caspase-9) pathway and the extrinsic pathway (Fas- associated protein with death domain, cleaved caspase-8, cleaved caspase-3, and cleaved poly adenosine diphosphate-ribose polymerase). These data suggest that PGG might be a potential therapeutic compound for melanoma.
